Effect of Sialadenectomy or Administration of Epidermal Growth Factor on Initiation of Hepatoma in Rat


Abstract
This study was performed to elucidate the modifying effect of epidermal growth factor (EGF), one of the potent initiator of cell division, on the rat hepatoma development. The SoltFarber's hepatocarcicogenesis model by administration of
diethylnitrosamine, acetoaminofluorene and partial hepatectomy was performed on Spragus-Dawley male rats. Administration of EGF was performed via subcutaneous injection of urogastron(5 mg/animal) three times a week during three weeks. Bilateral
sialadenectomy was done to eliminate the secretion of EGF from the salivary gland. Preneoplastic lesion was monitored by immunohistochemical method using the antibody against placental form of glutathione transferase. The result showed that there
was no
difference in development of enzyme-altered foci regardless of sialadenectomy or EGF aministration. Although a possibility of false negative result may remain, EGF seems to influence ittle effect in modification of the emengence of enzyme altered
foci
in Solt-Farber rat hopatoma model.
